BackgroundInformation | Dopaminergicneurondysfunctionandlosscorrelateswithnumerousprogressiveanddevastatingneurologicaldiseases.Stemcellresearchandapplicationsarerapidlymovingtowardtheintroductionandsubsequentpost-developmentanalysisofembryonicstem(ES)cell-deriveddopaminergicneuronsinadulttissue.TheNS145ispartofanexpandingChemicon/UpstateproductlinetargetingdevelopmentandplasticityinneuroBIOLOGy.NS145isanovel,broadlyencompassingkitforthelocalization,characterizationandanalysisofEScellderiveddopaminergicneuronintegrationintoadultcentralnervoussystem(CNS)tissue.Thiskitallowsresearchersto:(1)identifyandcharacterizedopaminergicneuronsintissue;and(2)identifyotherneighboringneuronaltypesandastrocytes.Inaddition,theantibodiesareamixtureofmouseandratmonoclonalsandrABItandgoatpolyclonalstooptimizedouble,triple,andquadruplestainingparADIgms.Thekitcomponentbreakdownisasfollows:
(1)Identifyingdopaminergicneurons.ResearchersareinterestedinidentifyingthesubtypeofneuronthattheirtaggedEShavematuredintofollowingintroductionandintegrationintotherecipientCNS.Thisisakeystepindopaminergicneuronreplacementtherapy.ThiskitincludesantibodiesfortheidentificationofdopaminergicneuronsusingseveraldiverseMarkerstargetingdifferentstagesintheneurotransmittersynthesisandmaintainance:DopaDecarboxylase(AB1569-50UL);TyrosineHydroxylase(TH;MAB5280-20UG);DopamineTransporter(DAT;MAB369-50UL).
(2)Identifyotherneuronalsubtypesandastrocytesneighboringthetransplantedcells.InordertomeasurethelevelofstructuralintegrationofthetaggedEScells,researchersoftenusegeneralmarkersofneuronalsubtypeandastrocytestocharacterizethelocalenvironmentinwhichtheEScell-deriveddopaminergicneuronsareintegrating.Thekitincludesantibodiesforthegeneralidentificationofneurons(NeuN;MAB377-100UG)andastrocytes(GFAP;AB5804).Alsoincludedareexcellentmarkersfortheidentificationofcholinergic(ChAT;AB144P-200UL)andSEROtonergic(Serotonin;MAB352-50UL)neurons.
