BackgroundInformation | PEA-15isa15kDacytoplasmicproteinwhichisabundantlyexpressedinastrocytes,althoughithasbeendetectedinavarietyofothercellsincludingthoseoftheheartandmuscle.ItmaybephosphorylatedonSer104byproteinkinaseCandonSer116byCAMKII,inacalcium-dependentmanner,inresponsetovarioushormonesandneurotransmitters.PEA-15alsocontainsanN-terminaldeatheffectordomain(DED)whichischaracteristicofproteinsthatregulateapoptosis.PreviousstudieshavereportedthatitinhibitsapoptosisinducedbythesignalingofTNFαinastrocytes.However,itisbecomingincreasinglyclearthatPEA-15isamultifunctionalproteinaffectingdiversecellularprocesses,possIBLybyitsmodulationoftheMAPkinasesignalingpathway.PreviousstudieshavedemonstratedthatPEA-15bindstoERKkinaseinthecytoplasm,andpreventstheactivationandnucleartranslocationofERK.Consequently,PEA-15blockstranscriptionaleventsandcellularprocessesmediatedbyERKanditsnucleartargetElk-1. |
