LargeCellColumnsaredesignedforpositiveselectionoflargehumanoranimalcells,forexamplemegakaryocytes.Ingeneral,cells
USPensionsshouldnotcontainclumps,aggregatesorparticles>50μm.Theflowrateofthecolumnsiscontrolledwithaflowresistorthatissuppliedwiththecolumns.

ThematrixoftheLargeCellColumnsiscomposedofferromagneticspheres,whicharecoveredwithacell-friendlycoatingallowingfastandgentleseparationofcells.WhenplacedinthemagneticfieldofaMACS

ADIentwithinthecolumn.ThisiscrucialforisolationofcellswhichareonlyminimallylabeledwithMACS
?MicroBeads,leavingenoughepitopesfreeforconcurrentantibodystaining.Thespacebetweenthespheresisseveraltimeslargerthanprimaryandmostculturedcells.Thisallowsthecellstofreelyflowthroughthecolumn.Magneticallylabeledcellsareheldinsuspensionwithinthecolumnanddonotactually“bind”thecolumnmatrix.Thissuspensionminimizesstressonthecellsandallowsforefficientsterilewashingbyavoidingcellaggregation.
